Rhouma, A., Boubaker, A., Nesme, X., and Dessaux, Y. 2006. Plasmid and Chromosomal Diversity of a Tunisian Collection of Agrobacterium tumefaciens Strains. Tunisian Journal of Plant Protection 1: 73-84. Biochemical and molecular characteristics of Agrobacterium tumefaciens strains isolated in Tunisia from different host plants were analysed. A collection of 109 isolates was obtained from diseased plants in several nurseries and orchards located in different regions. The majority of strains was isolated from stone fruit trees and belong to the biovar 1. The analysis of opines produced in tumors revealed that the local strain plasmids were of nopaline type. The 16S rRNA gene sequencing showed the presence of different genomic species among the Tunisian collection; but analysis was not enough to undoubtedly assign any isolate to a single genomic species. The multi-loci analysis carried out by BOX-PCR showed that the local isolates were assigned to frequent genomic species G4 (related to the reference strain B6) and G1 (related to reference strain TTT111), but also to rare genomic species G6 and G7 related respectively to Zutra F/1 and RV3.

Molecular identification of agrobacterium tumefaciens containing pCAMBIA 1305.2 plasmid using multiplex PCR and Gold nanoparticles multiplex probe

Conventional microbiology methods used to detect bacteria include multiple cultures and identification processes, so the results of lab work are painstaking and time-consuming. In recent years, more and more tend to use the diagnostic tests which are based on DNA; hence, DNA diagnostic biosensors have been created to perform DNA identification better. In this study, GUS and hpt genes were used ...

Plasmid-dependent attachment of Agrobacterium tumefaciens to plant tissue culture cells.

Kinetic, microscopic, and biochemical studies show that virulent Ti (tumor inducing)-plasmid-containing strains of Agrobacterium attach to normal tobacco and carrot tissue culture cells. Kinetic studies showed that virulent strains of A. tumefaciens attach to the plant tissue culture cells in increasing numbers during the first 1 to 2 h of incubation of the bacteria with the plant cells. Five T...

Genome analysis of Agrobacterium tumefaciens: construction of physical maps for linear and circular chromosomal DNAs, determination of copy number ratio and mapping of chromosomal virulence genes.

The phytopathogenic bacterium Agrobacterium tumefaciens is unique in that it possesses both linear and circular DNA chromosomes in addition to a plant-tumor-inducing (Ti) plasmid. We analyzed the two chromosomal DNA molecules in strain MAFF301001, whose Ti plasmid has already been sequenced completely. Physical maps of the chromosomal DNAs were constructed by Southern hybridization experiments ...

Virulence of Agrobacterium tumefaciens Strain A281 on Legumes.

This study addresses the basis of host range on legumes of Agrobacterium tumefaciens strain A281, an l,l-succinamopine strain. We tested virulence of T-DNA and vir region constructs from this tumor-inducing (Ti) plasmid with complementary Ti plasmid regions from heterologous nopaline and octopine strains.
